Mechanical cues play a role in cancer metastasis

Claude Verdier

We investigate some mechanical cues of the metastatic cascade, related to the migration of cells escaping from tumors, and migrating through tissues, or through the endothelial wall of blood vessels. Several tools have been used, in particular Traction Force Microscopy (TFM) for the follow-up of time-dependent stresses during the in vitromigration of cancer cells on specific gels (Peschetola et al. 2013), but also AFM measurements showing the interactions between cancer cells and endothelial cells (Laurent et al. 2014). By doing this, we obtain information on receptors and ligands involved during such interactions. Finally, using an AFM in force modulation mode, we measure the micro-rheological properties (Abidine et al., 2014) of more or less invasive cancer cells (epithelial bladder cancer lines). Altogether these results give new insights into the understanding of the metastatic process.
